Genotype analysis of HIV-1 gag genes from cases diagnosed as men who have sex with men in Harbin

Abstract:

Objective

To investigate the gag genes derived from cases diagnosed as HIV-1-infected men who have sex with men (MSM) in Harbin.

Methods

Genomic DNA were extracted from peripheral blood mononuclear cells (PBMCs) of 49 MSM with HIV-1-infection diagnosed as HIV-1 positive. The gag gene fragments were amplified by nested PCR. Phylogenetic tree was constructed to observe gag clustering and subsequently to analyze the genotyping feature of these cases.

Results

Total of 49 sequences of gag gene were obtained successfully. The phylogenetic analysis showed that these sequences clustered with three subtype lineages as follows: 39 sequences (79.6%) clustered with CRF01_AE lineage, 5 sequences (10.2%) clustered with subtype B, 4 sequences (8.2%) clustered with CRF07_BC. Moreover, one recombinant genotype CRF01B (2.0%) was also identified. And 1 sequence (2.0%) clustered with subtype CRF01B. Among CRF01_AE sequences, 92.3% (36/39) gathered with reference genes from Chinese MSM, and 7.7% (3/39) gathered with reference genes epidemic in Thailand and Southwest regions of China.

Conclusions

The CRF01_AE was the predominant genotype among cases diagnosed as HIV-1-infected MSM in Harbin. The MSM population may become the important source of new genotypes and complicated recombinant forms. Therefore, timely epidemiological surveillance of HIV-1 molecular epidemiology and development of prevention measures for HIV-1 infections among MSM are important.
